Neurodegenerative diseases such as Alzheimer’s disease, Parkinson’s disease, frontotemporal dementia, and dementia with Lewy bodies often present with overlapping clinical features and mixed underlying pathology, limiting diagnostic precision. This presentation describes a large-scale proteomics and artificial intelligence framework developed to classify major neurodegenerative diseases and quantify co-pathology at the individual level using cerebrospinal fluid and plasma.
Leveraging proteomic data from more than 24,000 samples across multiple independent cohorts, the approach demonstrated strong performance in distinguishing major disease categories, generalized across external datasets, and showed transferability across assay formats. We will highlight how high-dimensional protein profiling enabled identification of disease-relevant signatures, alignment with neuropathologic measures, and detection of mixed pathology in clinically ambiguous cases.
The session will discuss the potential of scalable blood- and CSF-based proteomics to improve disease characterization beyond traditional biomarkers and to support patient stratification, longitudinal monitoring, and precision-medicine approaches in heterogeneous and preclinical neurodegenerative disease populations.
